چکیده

Monoclonal anti-LDL antibodies were produced in a mouse spleen-myeloma system and purified by affinity chromatography on insolubilized low density lipoprotein (LDL). Five antibodies with different specificities could be distinguished by their immunoreactivities with chemically modified LDL preparations, and by their competition for binding to LDL. One of the antibodies inhibited the binding of (125)I-labeled LDL to the apoB,E receptors of cultured human fibroblasts. The same degree of inhibition was achieved using isolated Fab fragments. This antibody may bind to an antigenic site located near the cellular binding site of LDL-apoB.-Tikkanen, M. J., R. Dargar, B. Pfleger, B. Gonen, J. M. Davie, and G. Schonfeld. Characterization of antigenic determinants of human apolipoprotein B. Distribution on tryptic fragments of low density lipoprotein.

In the hope of obtaining useful probes to study the structure of human apolipoprotein B (apo B), we characterized monoclonal antibodies against low density lipoprotein (LDL). We examined the distribution of their corresponding antigenic determinants on tryptic fragments of LDL separated by monodimensional (SDS) or two-dimensional electrophoresis. Each antibody reacted with several different fra...

Identification of a mouse monoclonal antibody, LHLP-1, specific for human Lp(a).

Heretofore, immunologic reagents used to define and quantify human Lp(a) have been polyclonal in origin and therefore heterogeneous in antigenic specificity. We report here the isolation of a mouse monoclonal antibody, LHLP-1, monospecific for Lp(a). The antigen reactive with LHLP-1 was expressed in both lipoprotein Lp(a) as well as apolipoprotein Lp(a) delipidated by SDS treatment; however, di...

Analysis of the antigenic composition of liver specific lipoprotein using murine monoclonal antibodies.

Eight monoclonal antibodies have been raised to antigenic determinants within the liver specific lipoprotein complex. Five of these determinants were species and the others non-species specific. No liver specific determinants were identified. Liver specific lipoprotein antigens recognised by the eight monoclonal antibodies were located either on the hepatocyte membrane and/or along the sinusoid...

Production and Partial Characterization of Monoclonal Antibodies to Leishmania

Background: Monoclonal antibody technology allows identification of amastigote-specific antigen in human tissue biopsies and search for a suitable vaccine candidate, which may induce a long lasting immunity. Objectives: Production and partial characterization of seven monoclonal antibodies against Leishmania tropica promstigotes. Methods: Seven murine monoclonal antibodies of the IgG isotype we...

Production and Characterization of Murine Monoclonal Antibodies Recognizing Conformational and Linear Epitopes Localized on Human IgA2 Molecules

Background: There are two subclasses of human IgA (IgA1 and IgA2) that differ in antigenic properties and in chemical composition. The constant domains of α1 and α2 heavy chains have >95% sequence homology though major structural differences exist in the hinge region. Quantitation of IgA subclass levels depends on the availability of monoclonal antibodies (MAbs) specific for conserved conformat...
